Cloning and Characterization of Nitrate Reductase Gene in Kelp Saccharina japonica (Laminariales, Phaeophyta)

Summary

  • Nitrate reductase (NR) is an important enzyme in the nitrogen cycle, and its activity is closely related to the growth and development of plants.
  • In this study, the NR gene from the kelp Saccharina japonica was cloned and characterized.
  • The full-length cDNA of the NR gene was 1,845 bp in length, and the open reading frame (ORF) was 1,717 bp, encoding a polypeptide of 572 amino acids.
  • The deduced amino acid sequence of the NR gene showed high similarity to those of other species.
  • The expression of the NR gene was highest in the sporophyte and lowest in the gametophyte.
  • The NR gene was induced by nitrate and repressed by ammonium.
  • The NR gene was also induced by abscisic acid and salicylic acid.
  • The NR gene was localized to the cytoplasm and nucleus.

Detailed Description

Nitrate reductase (NR) is an important enzyme in the nitrogen cycle, and its activity is closely related to the growth and development of plants. In this study, the NR gene from the kelp Saccharina japonica was cloned and characterized. The full-length cDNA of the NR gene was 1,845 bp in length, and the open reading frame (ORF) was 1,717 bp, encoding a polypeptide of 572 amino acids. The deduced amino acid sequence of the NR gene showed high similarity to those of other species. The expression of the NR gene was highest in the sporophyte and lowest in the gametophyte. The NR gene was induced by nitrate and repressed by ammonium. The NR gene was also induced by abscisic acid and salicylic acid. The NR gene was localized to the cytoplasm and nucleus.

The NR gene was cloned from the kelp Saccharina japonica, a species of brown algae in the Laminariales order of the Phaeophyta division. The full-length cDNA of the NR gene was 1,845 bp in length, and the open reading frame (ORF) was 1,717 bp, encoding a polypeptide of 572 amino acids. The deduced amino acid sequence of the NR gene showed high similarity to those of other species.

The expression of the NR gene was analyzed in different tissues of S. japonica. The results showed that the expression of the NR gene was highest in the sporophyte and lowest in the gametophyte. The NR gene was also induced by nitrate and repressed by ammonium. The NR gene was also induced by abscisic acid and salicylic acid.

The subcellular localization of the NR gene was also studied. The results showed that the NR gene was localized to the cytoplasm and nucleus. This suggests that the NR gene may play a role in both the cytoplasm and nucleus.

Conclusion

In conclusion, the NR gene from the kelp Saccharina japonica was cloned and characterized. These results provide valuable information for further studies on the role of the NR gene in the growth and development of S. japonica.
